Connexin 43/GJA1 [F7]
Description The connexins are a group of gap junction proteins which form a hexamer to compose a connexon. Clusters of connexons form a gap junction through which low molecular weight proteins may diffuse from cell to cell. Several mammalian cells with malignant phenotypes exhibit decreased connexin expression and gap junction communication. In Src transformed cells, there is a decrease in gap junctional communication, which appears to be associated with tyrosine phosphorylation of connexin 43. Activated c-Src phosphorylates the C-terminal tail of connexin 43 on Tyr 265, resulting in a stable interaction between both proteins, which leads to inhibition of gap junctional communication. In addition to tyrosine phosphorylation, connexin 43 has also been shown to be phosphorylated on serine in the absence of Src kinases and on both serine and tyrosine in cells expressing Src kinases, such as c-Src and/or pp60v-Src. In human vascular endothelial cells, connexin 43 is posttranslationally modified during Host Mouse Application Immunofluorescence (IF), Immunohistochemistry (IHC), Immunoprecipitation (IP), Western Blot (WB) Reactivity Human, Mouse, Rat, Bovine -
Cytokeratin 8 [35BH11]
Description Cytokeratin 8 (CK8) is an intermediate filament protein produced early in embryogenesis. It is the only type-II CK occurring in many simple epithelial in respiratory, gastrointestinal, male and female reproductive tract and thyroid. CK8 is often co-expressed with Cytokeratin 18. CK8/18 is the major keratin pair in simple-type epithelia, as found in the liver, pancreas, and intestine. CK8 is used to detect adenocarcinomas with simple epithelium origin. The difference in staining pattern is useful to distinguish duct (peripheral staining) from lobular (perinuclear staining) breast carcinoma. FDC/Follicular dendritic cell [CNA42]
Description The monoclonal antibody CNA.42 recognizes a unique carbohydrate epitope found on follicular dendritic cells (FDCs) from a variety of species. FDCs are large cells in germinal centers (GCs) of primary and lymphoid tissues. They present non-processed antigen and switch off apoptosis of B cells through a selection process resulting in selection of memory B cells. FDCs express many B cell markers such as CD21 (long isoform), CD23 and CD35. A role for FDCs as been shown in various infectious diseases such as AIDS and transmissible spongiform encephalitis. Staining with this CNA.42 monoclonal antibody is mainly restricted to the plasma membrane but in limited cases may also be cytoplasmic (as seen in mast cells). Hairy Cell Leukemia [DBA.44]
Description Hairy cell leukemia (HCL) is a rare type of chronic lymphoid leukemia. The disease is characterized by abnormal white blood cells bearing hair-like projections from the cytoplasm. These cancerous cells are larger than normal and positive for CD19, CD20, CD22, CD11c, CD25, CD103 and FMC7. HCL commonly causes infection, anemia and/or easy bleeding in patients. Some of the leukemic cells may gather in the spleen and cause it to swell, leading to massive splenomegaly. Patients with a high tumor burden may also have significantly reduced levels of cholesterol. There are two variants of hairy cell leukemia: hairy cell leukemia-variant, which is usually diagnosed in older men; and a Japanese variant. HCL markers are important research tools as they allow for the functional and behavioral analysis of this type of leukemia. DBA.44, a B subset antibody, reacts with very few normal cells. 50-97% of HCL cases are positive; about 35% of low grade B-cell lymphomas are positive; and about 30-80% of s Host Mouse Application Immunohistochemistry (IHC) Reactivity Human -

IPO-38 (Proliferation Marker) [IPO38]
Description Recognizes a protein of 14-16kDa, which is a novel nuclear antigen of proliferating cells. IPO-38 antigen is present in the nuclei of proliferating cells such as Hodgkin s disease and non-Hodgkin s lymphomas, different forms of leukemias, breast and colorectal carcinomas, and PHA-stimulated lymphocytes. It is not expressed in the cells of non-stimulated lymphocytes and granulocytes. IPO-38 may be a useful marker of cell proliferation during monitoring of tumor progression. Laminin Receptor/RPSA [MD164]
Description Laminins, a family of extracellular matrix glycoproteins, are the major non-collagenous constituent of basement membranes. They have been implicated in a wide variety of biological processes including cell adhesion, differentiation, migration, signaling, neurite outgrowth and metastasis. Many of the effects of laminin are mediated through interactions with cell surface receptors. These receptors include members of the integrin family, as well as non-integrin laminin-binding proteins. This gene encodes a high-affinity, non-integrin family, laminin receptor 1. Reportedly, level of laminin receptor transcript is higher in colon carcinoma tissue and lung cancer cell line than their normal counterparts. Also, there is a correlation between the upregulation of this polypeptide in cancer cells and their invasive and metastatic phenotype. Macrophage Specific [LN-5]
Description LN5 stains an unidentified antigen in cytoplasm of macrophages and histiocytes in hematopoietic organs. It stains mantle zone B lymphocytes of the lymph node and spleen, spermatogonia, chief cells of the stomach, ductal epithelium of breast and tubular epithelium of kidney. It is strongly reactive with cases of true histiocytic lymphoma but is negative, except for macrophages, in Hodgkin's disease and non-Hodgkin's lymphomas. It can be an important tool for the study of malignant and benign histiocytic lesions.
