Ornithine Decarboxylase-1 (ODC-1) [ODC1/485]
Description Belongs to the Orn/Lys/Arg decarboxylase class-II family. Recognizes a 53kDa protein, identified as the Ornithine Decarboxylase (ODC-1). ODC is the initial and rate-limiting enzyme in the biosynthetic pathway of polyamines and is involved in the conversion of ornithine to putrescine. The biological activity of ODC-1 is rapidly induced in response to virtually all agents known to promote cell proliferation including hormones, drugs, growth factors, mitogens, and tumor promoters. Reportedly, ODC mRNA levels are elevated in lung carcinomas as well as in colon adenomas and carcinomas. ODC activity in colorectal carcinomas is greater than those in adenomas and normal mucosa. Osteocalcin/BGLAP [G5]
Description Bone γ-carboxyglutamic acid (Gla) protein, known as BGLAP, BGP or osteocalcin, is an abundant, non-collagenous protein component of bone that is produced by osteoblasts. In mice, osteocalcin is composed of a cluster of 3 genes known as OG1, OG2 and ORG, all of which can be found within a 23 Kb span of genomic DNA. Human osteocalcin is a highly conserved, 46-50 amino acid, single chain protein that contains 3 vitamin K-dependent γ-carboxyglutamic acid residues. Osteocalcin appears transiently in embryonic bone at the time of mineral deposition, where it binds to hydroxyapatite in a calcium-dependent manner. In addition, osteocalcin is one of the most abundant, non-collagenous proteins found in mineralized adult bone. Genetic variation at the osteocalcin locus on chromosome 1q22 impacts postmenopause bone mineral density (BMD) levels and may predispose some women to osteoporosis. OTK4/FKBP1B/FKBP12.6 [H8]
Description Immunophilins are a highly conserved family of cis-trans peptidyl-prolyl isomerases which bind to and mediate the effects of immunosuppressive drugs such as Cyclosporin, FK506 and Rapamycin. FKBP12.6, also known as FK506-binding protein 1B, is a 108 amino acid immunophilin belonging to the FKBPtype PPIase family. Subcellularly localized to the cytoplasm, FKBP12.6 binds to RyR in cardiac muscle sarcoplasmic reticulum and possibly plays a unique physiological role in excitation-contraction coupling in cardiac muscle. FKBP12.6 also catalyzes the cis-trans isomerization of proline imidic peptide bonds in oligopeptides. Ubiquitously expressed, FKBP12.6 is found at highest levels in brain and thymus. FKBP12.6 is expressed as two isoforms produced by alternative splicing. p27/Kip1 [DCS-72.F6]
Description p27Kip1 is a cyclin-dependent kinase inhibitor involved in G1 arrest. p27Kip1 binds to and inhibits cyclinE-Cdk2 complex, cyclinA-CDK2 and cyclinD1-CDK4 (1). p27Kip1 is regulated by phosphorylation on serine 10 (S10) and threonine 187 (T187). Phosphorylation by CDK2 on T187 results in ubiquitination and degradation of p27Kip1, while phosphorylation by hKIS on S10 signals nuclear export to the cytoplasm. The expression level of p27Kip1 is high in normal cells. Downregulation of p27Kip1 is found in many types of cancers, and decreased expression of p27Kip1 appears to be a poor prognostic factor in several tumor models, including carcinomas of the lung, breast, colorectal, and prostate. p34/CDC2/CDK1 [POH-1]
Description p34cdc2 is a phosphoprotein with protein kinase activity that functions in the G2/M phase transition of the cell cycle. It is the catalytic subunit of the maturation-promoting factor (MPF) and complexes with both cyclin A and B in mammalian cells. Activated p34cdc2 kinase phosphorylates a variety of substrates leading to some specific events of mitosis including nuclear envelope break-down and chromosome condensation. p34cdc2 has also been implicated in lymphoid proliferation. p40 Polyclonal
Description p63 consists of two major isoforms-TAp63 and ΔNp63. These isoforms differ in the structure of the Nterminal domains. The TAp63 isoform identified by anti-p63 antibody) contains a transactivation-competent ‘TA’ domain with homology to p53, which regulates the expression of the growth-inhibitory genes. In contrast, ΔNp63 isoform (identified by anti-p40 antibody) contains an alternative transcriptionally-inactive ‘ΔN’ domain, which antagonizes the activity of TAp63 and p53. The p40 (clone ZR8) antibody recognizes exclusively ΔNp63 but not TAp63. p40 is a squamous cell carcinoma ‘specific’ antibody. It reacts with the vast majority of cases of squamous cell carcinomas of various origins, but not with adenocarcinomas. It is particularly useful in differentiating lung squamous cell carcinoma from lung poorly differentiated denocarcinoma. p40 antibody can also be used as an alternative basal cell/myoepithelial cell marker, which has similar sensitivity and specificity as that of p63 antibody.
